Chapter 3 Immiscible Langmuir monolayers: Interactions between the polar head groups of cholesterol and octylcyanobiphenyl

چکیده

Mixed Langmuir monolayers are ideal systems to mimic biological membranes [1, 2]. Studies on mixed monolayers provide information about the interactions involved, miscibility and stability of the monolayer. It is known that cholesterol(Ch) is found in bilayer membranes and lipoproteins. Ch regulates the transport and barrier properties of the membranes [3]. Lung surfactant in the human body is composed of different kinds of lipids and proteins. This forms Langmuir monolayer at the alveolar air-water interface [4, 5, 6]. These mixed surfactants tend to reduce the surface tension to nearly zero. The mixed monolayer is required to be stable without going to the collapsed state during expiration. Lack or deficiency of these surfactants lead to respiratory distress syndrome(RDS) in the human body. Recent studies on mixtures of saturated lipid, unsaturated lipid with different concentration of cholesterol to mimic raft formation in membranes bring in some correlation between monolayer and unilamellar vesicles [7]. There are many reports on the mixed monolayer of cholesterol with biologically active lipids [8]. The mixed monolayer of cholesterol and oleic acid shows good miscibility [9]. This is attributed to Ch filling the voids in the bent shape molecule, oleic acid. With increase

Polar head group interactions in mixed Langmuir monolayers.

We have investigated the miscibility in the mixed monolayers of cholesterol (Ch)-octyl cyano biphenyl (8CB) and cholesteryl acetate (ChA)-8CB using surface manometry and epifluorescence microscopic techniques. The main skeleton is the same both in Ch and ChA, whereas the polar head group is alcohol in Ch and ester in ChA. The 8CB molecule has a polar cyano as a terminal group and we probe its i...

Interactions of gangliosides with phospholipids and glycosphingolipids in mixed monolayers.

1. The interactions among five different gangliosides and three chemically related glycosphingolipids and their behaviour in mixed monolayers with six different phospholipids were investigated at the air/145 mM-NaCl interface at pH 5.6. 2. The mixed monolayers of any of the different gangliosides showed an immiscible behaviour at high surface pressures, with absence of interactions among them r...

Interaction of pulmonary surfactant protein A with dipalmitoylphosphatidylcholine and cholesterol at the air/water interface.

Interaction of pulmonary surfactant protein A (SP-A) with pure and binary mixed dipalmitoylphosphatidylcholine (DPPC) and cholesterol (3.5 wt%) at the air/saline, 1.5 mM CaCl2 interface was investigated using a rhomboid surface balance at 37 degrees C. Surface tension-area isotherms were measured to access the surface active properties of the monolayers. The organization of DPPC and cholesterol...

Mixed monolayers of cholesteric acid and lipid molecules

The studies on Langmuir monolayer of cholesteric acid (ChA) at the air-water (A-W) interface show many interesting results. We have observed different two-dimensional (2D) phases like L′1, L ′ 2 and L2. In this chapter, we describe our studies on the stability of the phases and miscibility of binary mixture of ChA and lipid molecules of different structures. We have chosen the lipid molecules l...

Dipole potential and head-group spacing are determinants for the membrane partitioning of pregnanolone.

The membrane interactions of pregnanolone, an intravenous general anesthetic steroid, were characterized using fluorescence spectroscopy and monolayer technique. di-8-ANEPPS [4-[2-[6-(dioctylamino)-2-naphthalenyl]ethenyl]-1-(3-sulfopropyl)-pyridinium], a membrane dipole potential (Psi)-sensitive probe, revealed pregnanolone to decrease Psi similarly as reported previously for other anesthetics....
